Methods In total 85 individuals of S. baicalensis herbs were collected and t he surface soils under each of t he plant were also sampled f rom eight different habitat s, wit h an interval of about m vertically ranging f rom 800 to m in August t hrough September, The content s of active component s in the herbal samples, namely baicalin, wogonoside, baicalein, wogonin, and oroxylin A were determined by HPL C. The organic matter and t he total nitrogen co ntent s in t he soil samples were analyzed t hro ugh standard measurement p rocess. The vario us habitat conditions represented by temperat ure were estimated t hrough a vertical lap se rate of / 100 m, and t hose by precipitation and solar radiation calculated according to t he isoline map s. Results The signi2 ficant positive correlations were observed only between the content s of baicalin, wogonoside and the eleva2 tion. Bot h t hose two content s showed the significant negative correlations wit h t he annual mean tempera2 ture and precipitation, while showed positive wit h annual mean solar radiation. No coorelation wit h any statistical significance was observed between t he content s of active component s in S. baicalensis and the soil parameters in t his st udy. Conclusion It seems t hat t he plant s of S. baicalensis growing in a habitat wit h annual mean temperat ure wit hin a range of 2 6 could concentrate relatively rich content s of active co mpo nent s. Wit hin t he nat ural dist ributio n areas of S. baicalensis, t he habitat s wit h an annual tem2 perat ure averaged at 3 4 tend to be t he most fitting sites for growing best quality S. baicalensis rich in active component s, and t ho se sites are almost coincident wit h t he environment for trueborn production area of t raditio nal S. baicalensis herbs. 3 : : (2006BAI09B0225) ; ( ) :(1982 ),,,, Tel : (010) E2mail : cn 3 E2mail : cn

Zhejiang Research Institute of Chinese Materia Medica, Hangzhou , China) Abstract : Objective The A FL P analysis of nine accessions of L eonurus j a ponicus was conducted to explore genetic diversities among germplasm. Methods Genetic diversities at DNA level among t he germ2 plasm of L. j a ponicus were studied by t he amplified f ragment length polymorp hism (A FL P) marker, and 32 pair s of selective amplificatio n p rimer were identified. The similarities amo ng t he germplasm of L. j a ponicus were calculated and analyzed by t he sof t ware of N TS YS2PC, and genetic evolutio n t ree was developed, using amplified f ragment s as p rimary matrix. Results The quality of genomic DNA of L. j a ponicus ext racted by SDS met ho d was good eno ugh to meet t he requirement of A FL P analysis. Ten combinations wit h multiple morp hologies, clear bands, and high discrimination rate were identified f rom 32 pairs of selective amplification primers. The A FL P fingerprinting was developed, which could differen2 tiate each of nine accessions of L. j a ponicus. Nine accessions of L. j a ponicus could be classified into two types based on t he evolution tree.
