MBeth, David mersn* Bigelw Labratry fr ean Sienes, ast Bthbay, M SAjmbeth@bigelw.rg, demersn@bigelw.rg ('presenting athr) Little is knwn abt the mirbial elgy f rrding steel in marine envirnments 1-2 r f the natral abndane f irn-xidizing bateria (FeB) in these systems. We hypthesized that astal sediments are reservirs fr the marine FeB 'Zetaprtebateria' (Zetas), and that they an lnize and beme nmerially abndant n mild steel srfaes. A 40 day time series inbatin was ndted in a salt marsh (smmer 2010). rrsin mmnity DNA was extrated and analyzed fr baterial diversity with tagged pyrseqening (V4 regin, 16S rrna gene). Several relevant mmnities were qantified sing qpr: bateria and arhaea 3 and Zetas 4 sing 16S rrna gene speifi primers, and slfate-reding bateria (SRB) sing a dsra gene speifi primer 5. The pyrseqening data shwed the presene f Zetas in sediments and thrght the inbatins n the steel samples. Irn xyhydrxide stalk bisignatres were bserved n samples, frther evidene that these seqenes likely represent FeB. Relatives f the H2-xidizing gens Hydrgenphaga and members f the family Rhdbaterales were als identified as imprtant members f the birrsin mmnity and were present bth n steel and in sediments. Gene pies assessed with qpr remained fairly nstant in sediments dring the stdy, and Zetas were a 10-fld lwer than SRB. Zetas lnizing the steel inreased rapidly ver the first 10 days, exeeding pies qantified in the sediment by an rder f magnitde. The SRB nmbers n the steel were 10 fld lwer than in sediments dring the first days f inbatin, bt inreased t near the sediment levels by 40 days. This wrk illstrates that astal sediments are a reservir fr Zetas wh, thgh nmerially lw in sediments, an qikly lnize envirnments where free Fe(II) is abndant Referenes: (1) MBeth JM etal (2011) Appl nv Mirbil 77: ; (2) Dang H etal (2011) nv Mir 13: ; (3) Takai K & Hrikshi K (2000) Appl nv Mirbil 66: ; (4) Kat S etal (2009) nv Mirbil 11: ; (5) Ben- Dv etal (2007) Mirb l 54: MI-4 MARIN MI F MILD STL - LTRHMIAL ANALYSIS F MDL RRSIN MMNITIS Jye M. The bjetive f this wrk was t determine the eletrhemistry f mild steel hallenged with single strains f FeB and FeRB vs ltres f FeB and FeRB. Bath experiments ntaining mild steel pns in marine medim were ndted in green flasks. Pre and mixed ltres f marine FeB (Mariprfünds ferrxydans strain M34) 3 and FeB (Gethermbater sp. strain HR-1) 4 were sed in eah system, and ntrls ntaining n added FeB and FeRB were als prepared. Pre FeB were grwn in an aerbi blk medim and pre FeRB were grwn nder anaerbi nditins. rrsin rates were mnitred eletrhemially, and fllwing inbatin, steel srfaes were evalated with SM and prfilmetry. An FeB and FeRB -ltre was sessflly grwn in an blk aerbi envirnment, and the FeB-generated irn xide stalks in this treatment appeared dended in mparisn with thse frmed in the pre FeB system. Prfilmetry demnstrated less nifrm rrsin attak in the presene f FeB and FeRB -ltre mpared t all ther expsres. letrhemially mnitred plarizatin resistane sggested that all aerbi rrsin rates were similar and rders-f-magnitde higher than anaerbi rrsin rates.
