Furthermore, upon additional passage in HeLa cells the G64T mutation reverted to an alanine residue (GCC codon).

Poliovirus cdna clones encoding 3 poly(a) tails 32, 51 and 84 bases in length were transcribed by T7 DNA-dependent RNA polymerase into viral RNAs as described in Materials and Methods (Steil et al., 2010). An RNA linker was added to the 3 end of the viral RNA transcripts and a cdna primer complementary to the RNA linker was used in conjunction with Superscript III reverse transcriptase to make cdna. cdnas were amplified by PCR and TOPO-TA cloned into plasmids in E. coli.
