Waithe et al Supplementary Figures Supplementary Figure 1 Expression and properties of WT and W391A mutant YFP- Ca V 2.2. A Immunoblot using Ca V 2.2 Ab for untransfected cells (UT, lane 1), YFP-Ca V 2.2 (lane 2) and CFP-Ca V 2.2 (lane 3), showing protein bands of the expected molecular weight (~280 kda), indicated by arrow. B Immunoblot using GFP Ab for untransfected cells (UT, lane 1), free ecfp (lane 2), CFP- Ca V 2.2 (lane 3) and YFP-Ca V 2.2 (lane 4), showing free CFP of the expected molecular weight (~31 kda) only in lane 2, indicated by arrow. C I-V relationship for YFP-Ca V 2.2(WT) (, n = 8) and YFP-Ca V 2.2(W391A) (, n = 9), coexpressed with α 2 δ-1 and β1b. Data were fit with a modified Boltzmann equation, with V 50, act of -17.8 mv and -2.9 mv, and G max of 62 and 27 µs, for YFP-Ca V 2.2(WT) and YFP- Ca V 2.2(W391A), respectively. Recordings were made with 10 mm Ba 2+ in Xenopus oocytes. D Bar chart of I Ba at 0 mv, expressed as a % of the value for the WT construct, for YFP- Ca V 2.2(WT) (black bar, n = 8) and YFP-Ca V 2.2(W391A) (white bar, n = 9), recorded alone, or together (gray bar, n = 10). The statistical significances of the differences compared to control Ca V 2.2 were determined by one way ANOVA and Bonferroni s post-test. ** P<0.01. E Steady-state inactivation curves for YFP-Ca V 2.2(WT) (, n = 5) and YFP-Ca V 2.2(W391A) (, n = 4), from the same experiments as in (C), fit with a Boltzmann equation, with V 50, inact of - 67.2 mv and -49.8 mv, respectively. 1

A Immunoblot using Ca V 2.2 Ab (upper panel) and with GAPDH as a loading control (lower panel), for untransfected tsa-201 cells (UT, lane 1), cells transfected with YFP-Ca V 2.2/α 2 δ- 1/β1b (lane 2), YFP-Ca V 2.2/α 2 δ-1 (lane 3), YFP-Ca V 2.2(W391A)/α 2 δ-1/β1b (lane 4) and YFP- Ca V 2.2(W391A)/α 2 δ-1 (lane 5). Representative of three experiments with similar results. B Bar chart showing expression of Ca V 2.2 in tsa-201 cells, from 3 experiments including that illustrated in A, for YFP-Ca V 2.2(WT)/α 2 δ-1 (back and white bars) and YFP-Ca V 2.2(W391A)/α 2 δ- 1 (gray and hatched bars) either co-expressed with β1b (black and gray bars) or with empty vector (white and hatched bars). The data were normalized using GAPDH expression. N=3, with individual values plotted as open squares. * P<0.05, 1 way repeated measures ANOVA. C Bar chart of total neurite fluorescence density, for YFP-Ca V 2.2(WT) (black and white bars, n = 12, 13) and YFP-Ca V 2.2(W391A) (gray and hatched bars, n = 23, 9), either with the normal concentration of β1b (black and gray bars), or with β1b diluted 20-fold (white and hatched bars). Cells were injected after 24 h in culture and imaged 24 h later. The statistical significance between the different conditions is indicated by * P<0.05, ***P<0.001, 1-way ANOVA and Tukey s post-hoc test. 2

Supplementary Figure 3 Localization of ER and Golgi markers in SCG somata and growth cones. A Images of an SCG soma showing the expression of GFP-Ca V 2.2(WT) (left) compared with the distribution of the subcellular organelle marker, dsred-er (center). The merged image is shown on the right. The scale bars are 10 µm. B Images showing (left) the distribution of the GFP-Ca V 2.2 in a neurite, in comparison to the subcellular organelle markers dsred-golgi (center) in SCG growth cones. The merged image is shown on the right, and the extremities of the growth cones are identified by a dotted white line. The scale bar is 20 µm. C Bar chart of dsred-er fluorescence from data such as that in (Fig. 5A), for growth cones coexpressing GFP-Ca V 2.2(WT) (black bar, n = 20) and GFP-Ca V 2.2(W391A) (white bar, n = 21). There was no statistical significance between the two conditions (Student s t test). 3

Supplementary Figure 4 Retrogradely moving particles in neurites of SCG neurons expressing YFP-Cav2.2(WT). YFP-Cav2.2(WT) was expressed plus α 2 δ-1 and β1b in SCG neurons, and retrogradely moving particles were imaged in a region of interest indicated by the red box, 6 h after microinjection. Time-series were recorded in the YFP channel, and are provided as a separate movie (CAv2.2traffickingRetro5.avi). Scale bar 100µm. 4

Supplementary Figure 5 Effect of MG132 on total YFP+CFP-Ca V 2.2 fluorescence and on neurite outgrowth in SCG neurons. A Bar chart of the ratio of total YFP + CFP fluorescence in cell bodies, from data such as that in Fig. 7A, for YFP-Ca V 2.2(WT) (black bar, n = 14), YFP-Ca V 2.2(W391A) (white bar, n = 12), and YFP-Ca V 2.2(W391A) together with 50 nm (light gray bar, n = 13), 250 nm (dark gray bar, n = 13) or 1 µm (hatched bar, n = 13) MG132. All experiments also included CFP-Ca V 2.2(WT). The statistical significance between YFP-Ca V 2.2(W391A) in the absence and presence of MG132 is * P<0.05, 1-way ANOVA and Bonferroni s post-test. B Bar chart of the total neurite branch number for neurons expressing YFP-Ca V 2.2(WT) (black bar, n = 17), YFP-Ca V 2.2(W391A) (white bar, n = 17), and YFP-Ca V 2.2(W391A) together with 50 nm (light gray bar, n = 13), 250 nm (dark gray bar, n = 14) or 1 µm (hatched bar, n = 19) MG132. All experiments also included CFP-Ca V 2.2(WT), and are taken from experiments described in Fig. 7A-C. 5

Supplementary Figure 6 Controls for cell surface biotinylation and pull downs of Ca V 2.2(WT) and Ca V 2.2(W391A). A tsa-201 cells were transfected with Ca V 2.2/α 2 δ-1/β1b (lanes 1-4), or Ca V 2.2(W391A)/α 2 δ- 1/β1b (lanes 5-8). They were treated with either DMSO (vehicle control) or MG132 (250 nm) for 18 h. prior to harvesting, as indicated. Following cell-surface biotinylation, the biotinylated proteins were pulled down with streptavidin. The WCL and biotinylated fractions were blotted for Akt, to show that this intracellular protein was not biotinylated. This control was performed for every experiment shown in Fig. 8. B tsa-201 cells were transfected with Ca V 2.2/α 2 δ-1/β1b and either subjected to cell-surface biotinylation (lanes 3 and 4) or not (lanes 1 and 2), followed by streptavidin pull-down. Only the biotinylated cells showed a band for Ca V 2.2 following pull-down (lane 3), confirming the specificity of this procedure. 6

Supplementary Figure 7 Quantification of ubiquitinated Ca V 2.2(WT) and Ca V 2.2(W391A). A tsa-201 cells were transfected with YFP-Ca V 2.2/α 2 δ-1/β1b (lanes 1 and 2), or YFP- Ca V 2.2(W391A)/α 2 δ-1/β1b (lanes 3 and 4). They were treated with either DMSO (vehicle control) or MG132 (250 nm) for 18 h. prior to harvesting.
