10 p. 8 Applications PCP PCP Extraction of Phencyclidine from Urine and Analysis by GC/MS Phencyclidine (PCP) induces hallucinogenic and neurotoxic effects. Since a vast majority of the parent drug and its metabolite are excreted in urine, abuse is determined by detecting the parent drug. However, it is not structurally related to heroin or other opiate compounds. To determine abuse, a separate screening method must be performed. 12 p. 0 Applications THC THC Extraction of 9 Carboxy-THC from Urine and Analysis by LC/MS or GC/MS Tetrahydrocannabinol (THC), the main active chemical in marijuana, is mainly metabolized to 9-carboxy--or- 9 -carboxy-thc (THC-CH). This compound is then conjugated with glucuronic acid and excreted in urine. THC-CH and THC are effectively extracted from Strata-X-C and Screen-C through disruption of hydrophobicity and hydrogen bonding mechanisms. Removal of more polar matrix interferences can still be effectively done by washing with an organic solution acidified at or below ph. Due to the high volume of samples, system stability and fast run time are critical. Applications Co c a i n e Cocaine Extraction of Benzoylecgonine from Urine and Analysis by LC or GC/MS Cocaine is extensively metabolized by the liver, with only % being excreted in the urine unchanged. Abuse is thus detected by monitoring its main metabolite, benzoylecgonine. However, the ammonium hydroxide used for elution on cation-exchangers causes hydrolysis of the methyl ester in cocaine and cocaethylene converting them into carboxylic acid containing benzoylecgonine metabolite. Using a neutral sorbent such as Strata-X does not require high ph elution, allowing for independent analysis of all three cocaine compounds. 26 p. 4 Applications 6-MAM 6-MAM Extraction of 6-Monoacetylmorphine (6-MAM) from Urine and Analysis by GC/MS 6-Monoacetylmorphine is a unique metabolite of heroin and can be used to differentiate between users of heroin and other legal and illegal opiate drugs. SPE GC/MS Solutions Premier Solution: Strata -X or X-CW Low Cost Solution: Strata Screen-C Extraction of 6-MAM is similar to other opiate related species. A mixed-mode cation-exchange sorbent such as Screen-C or Strata-X-C ionically retains 6-MAM, and wash solutions with up to 00 % methanol are used to remove weakly bound matrix interferences. However, the use of ammonium hydroxide for elution can make the acetate group in 6-MAM unstable, converting it into morphine. To avoid any potential problems, use a weak cation exchanger such as Strata-X-CW or a neutral sorbent such as Strata-X. Strata-X-CW sorbent can be neutralized by eluting with % formic acid in methanol or any other oganic solvent.
