Poster D16 Q-bank An Online DNA Barcode Database For Identification Of Quarantine Organisms presented by Ewald (J.Z.) Groenewald

EPPO European and Mediterranean Plant Protection Organization Intergovernmental organization responsible for European cooperation in plant protection EU Council Directive for legislation A1 (absent) A2 (controlled) Total Fungi Bacteria & phytoplasmas Viruses & virus-like Insects & mites Nematodes Parasitic plants CBS-KNAW Fungal Biodiversity Centre

Problems Different names for the same or similar organisms: Dothistroma pini and Dothistroma septosporum: EPPO = Mycosphaerella pini EU Council Directive = Scirrhia pini Pseudocercospora pini-densiflorae: EPPO = Mycosphaerella gibsonii EU Council Directive = Cercoseptoria pini- densiflorae Frequently no cultures or reference material and sometimes no sequence data

Solution? Create an online reference database containing: taxonomy references to specimens (ex-type and or reference strains, preferably) link to quarantine information nucleotide sequences (preferably) morphology (if possible) disease symptom information (if possible) Database should cover the six main organism groups listed in EPPO

The Road to Q-bank FES project funded by Dutch government Approx Total budget EUR 9 million Fungi, Insects, Invasive Plants, Viruses & Viroids, (Bacteria, Nematodes) QBOL project from EU 7 th Framework Approx Total budget EUR 3 million Same as FES, but excluding Invasive Plants and including Phytoplasmas Curatorium funded by Dutch government Approx Total budget EUR 250 thousand 2014???

Q-bank Home page

Q-bank Bacteria page

Q-bank Bacteria page Barcode regions: 16S, GyrB, MutS, avrbs2

Q-bank Fungi page

Q-bank Fungi page Barcode regions: ITS, TEF, TUB (and/or COI and ACT for Phoma and Phytophthora)

Q-bank Insects page Barcode regions: COI (Lco-Hco primers) ITS

Q-bank Invasive Plants page Currently only morphology via a Lucid key

Q-bank Invasive Plants page

Q-bank Nematodes page Barcode regions: SSU, LSU, COI

Q-bank Phytoplasmas page Barcode regions: Tuf, 16S rdna

Q-bank Viruses page Barcode region: Blast against whole genomes

Q-bank Polyphasic Search pages YOU decide how much information you want to use in your identification; even sequence data from multiple loci!

Advantages of Q-bank Curated! Links to vouchered specimens For Fungi, links to MycoBank are provided Sequence data Links to EPPO and the EU Council Directive information General blast search as well as polyphasic identification Identification will lead you to correct Q status, irrespective of species name

Q-bank offers descriptions of well-characterised regulated plants pests comprises ecological, morphological, physiological, and sequence data of specimens available in physical collections serves as an identification and detection reference database for phytosanitary organisations and other end-users material used can be obtained for further studies or used as controls in identification and detection tests
