2 General: Reactions were performed under a nitrogen atmosphere in oven-dried glassware containing a tefloncoated stirrer bar and dry septum. Solvents were freshly distilled. GC analyses were carried out using a capillary column (Phenyl Methyl Siloxane 0 m x 0 x 0., 0/.-0-00/) and a time program beginning with min. at 0 C, followed by 0 C/min. ramp to 00 C, then min. at this temp. Column chromatography was performed using an MPLC system and RediSep packed column (1 g). TLC analyses were performed on commercial Kieselgel 0 F silica gel plates. MR spectra were obtained using CDCl or DMS-d as solvent, with proton and carbon resonances at 00 MHz or 00 MHz and 0 MHz or MHz, respectively. Synthesis of starting materials. Synthesis of -cyanobenzoic acid (c) [CAS: --]; The synthesis follows the literature procedure of T. Yoneta, S. Shibahara, S. Fukatsu, S. Seki, Bulletin of the Chemical Society of Japan 1, 1, -; and. Miyaura, Top. Curr. Chem., 1, Springer-Verlag, Berlin Heidelberg, 00, An oven-dried flask was charged with phthalic anhydride (. g, 1 mmol) and THF (0 ml). Ammonia was bubbled through the reaction solution for 1 min. under vigorous stirring at room temperature. The resulting white solid was filtered off, washed successively with small amounts of water and hot acetone, then dried under high vacuum to yield a white ammonium salt (1.1 g, %). This salt was dissolved in pyridine (00 ml) and stirred at room temperature for 0 min., then acetic anhydride (. g, 1. mmol) was added and the reaction mixture was stirred until a clear solution formed. The solvent was removed under reduced pressure, and the remaining solid was redissolved in ethyl acetate (00 ml). The organic phase was washed twice with aqueous HCl solution (1) and brine, and dried over MgS. The solvent was removed, and the solid was recrystallized from ethyl acetate to yield c as a white solid (1. g, %). 1H-MR (00 MHz, DMS-d ): = (m, 1H),.-. (m, 1H),.-. Synthesis of -(-bromophenyl)-1,-dioxolane (c) [CAS: 0-01-]; The synthesis follows the literature procedure of G. Drefahl, D. Lorenz, J. Prakt. Chem. 1,, An oven-dried flask, equipped with a dropping funnel filled with molecular sieves, was charged with -bromobenzaldehyde (. g, mmol), - toluenesulfonic acid (. mg, 0.0 mmol), ethan-1,-diol (1. g, mmol) and toluene (0 ml). The reaction mixture was refluxed over-night (1h), then cooled, washed successively with saturated aqueous ahc, and water, and dried over MgS. The toluene was distilled off. The crude yellow product was purified by Kugelrohrdistillation to yield compound c as a colorless liquid (.0 g, %). Anal. Calcd for C H Br : C,.; H,.. Found: C,.; H,.0. Synthesis of -cyano- -methylbiphenyl (a) [CAS: 11--1]; An oven-dried 0 ml crimp top vial was charged with copper(ii) oxide (11. mg, 0.1 mmol), potassium carbonate (11.1 mg, 0 mmol), potassium fluoride (.1 mg, 0.0 mmol), 1,-phenanthroline (.0 mg, 0.1 mmol), -cyanobenzoic acid ( mg, mmol), palladium(ii) bromide (. mg, 0.0 mmol), triphenylphosphine (. mg, 0.0 mmol) and ground Å molecular sieves (0 mg). The reaction vessel was closed with a septum cap, evacuated and flushed with nitrogen three times. Subsequently, a solution of -bromotoluene (1 mg, 0 mmol) and the internal standard n-tetradecane (0 µl) in quinoline (1. ml) was added via syringe. The resulting solution was stirred for h at C, then poured into aqueous HCl (1, 0 ml). The resulting mixture was extracted repeatedly with ethyl acetate (0 ml portions). The combined organic layers were washed with water and brine, dried over MgS, filtered, and the volatiles were removed in vacuo. An oven-dried 0 ml crimp top vial equipped with a septum cap was charged with copper carbonate (. mg, 0.0 mmol), potassium carbonate (1. mg, mmol), 1,-phenanthroline ( mg, 0. mmol) [these compounds were dried before for h at 0 C under high vacuum], -cyanobenzoic acid (1. mg, mmol), -bromobenzaldehyde ( mg, 0 mmol), palladium(ii) bromide (. mg, 0.0 mmol), triphenylphosphine (.0 mg, 0.0 mmol) and ground Å molecular sieves (0 mg, dried in the microwave and for h at C in high vacuum) and was flushed with nitrogen. 4 Large scale procedure for the synthesis -cyano- -methylbiphenyl (a) [CAS: 11--1]. An oven-dried flask was equipped with an internal thermometer, a dropping funnel filled with molecular sieves, a condenser, and charged with -cyanobenzoic acid (0. mg,.0 mmol), potassium carbonate (. mg,.0 mmol),. ml dry quinoline and. ml dry toluene. The reaction mixture was stirred for 1h at 0 C, refluxed for 0 min., then the toluene was removed in vacuo. After flushing the vessel with alternating vacuum and nitrogen purge cycles, copper(ii) oxide (. mg, 0. mmol), potassium fluoride (1. mg,.0 mmol) [these compounds were dried before for h at C under high vacuum], 1,-phenanthroline (1. mg, 0. mmol), -bromotoluene (1. mg,.0 mmol), palladium(ii) bromide (. mg, 0. mmol), triphenylphosphine (. mg, 0.0 mmol) were added. The vessel was lowered into an oil bath and heated to 10 C. The temperature inside remained constant at C. After stirring the reaction mixture for h, it was allowed to cool to room temperature. The solution was acidified with aqueous HCl solution (1, ml) and extracted repeatedly with ethyl acetate (0 ml portions). The combined organic layers were washed twice with aqueous HCL (1, ml), saturated ahc and brine, dried over MgS and filtered. The volatiles were removed in vacuo, and the residue was purified by Kugelrohr-distillation,) yielding a as a yellow solid (. mg, %). Large scale procedure for the synthesis -cyano- formylbiphenyl (b) [CAS: 1--]. 5 Representative procedure for the Valsartan synthesis -Bromomethyl--cyanobiphenyl (c) [CAS: 11--]; An oven-dried 0 ml crimp top vial equipped with a septum cap was charged with -cyano- -methylbiphenyl (1. mg, 0. mmol), -bromosuccinimide (. mg, 0. mmol), AIB (.0 mg, 0.01 mmol), dry cyclohexane (1. ml) and CCl ( ml). The reaction mixture was heated for 1h at C. The solvent was removed under reduced pressure and the crude product was purified by column chromatography (Si, ethyl acetate / hexane :). The resulting solid was recrystallized with drops ethyl acetate and hexane to yield compound c as white crystals (. mg, %). 1H-MR (00 MHz, CDCl ): =.0 (d, J =.Hz, 1H),..1 (m, 1H),.. (m, H),, (d, J =. The reaction mixture was stirred at room temperature for 0h. Afterwards, the solution was cooled to 0 C and sodium cyanoborohydride (. mg,.00 mmol) dissolved in ml EtH was added. The mixture was again stirred at C for h, then the molecular sieves were filtered off over Celite and the solution was washed with saturated aqueous ahc (ethyl acetate was added for a better separation) and dried over MgS. The reaction mixture was refluxed for h. After cooling to room temperature the solution was charged with 1. ml acetic acid (1 ml acid, ml water) and stirred over-night. The organic phase was separated, concentrated and purified by column chromatography (Si, DCM / (DCM, % MeH) = 1 / 1). The organic phase was separated and the aqueous layer was acidified with aqueous HCl solution () until a ph of was reached.
