Proposal Summary: The department of Chemistry and Biochemistry proposes the acquisition of a high performance liquid chromatograph with a diode array detection system (HPLC/DAD) for use in faculty and student research and in student instruction. This instrument will afford our students, particularly chemistry majors, with comprehensive hands-on experience in the use of a widely-used instrumental method for projects in various fields including, bioorganic, analytical and environmental chemistry and biochemistry.
2 Department of Chemistry and Biochemistry StAR Proposal Section I NARRATIVE HPLC is a robust, reliable technique with widespread applications. It is used extensively in many fields, including, but not limited to: (bio)pharmaceutical development and production; environmental monitoring and assessment; production and development of personal care products; assurance of consumer safety in the food and drug industries; and forensic investigations. The associated DAD allows for discrimination between structurally similar compounds, and is applied in areas such as drug safety (purity determination) and drug metabolism studies. The acquisition of an up-to-date HPLC system will serve two primary functions: it will be used in the research of one third of the faculty members in the department each of whom are currently at the Assistant Professor level. Secondly, it will serve as the focus for the development of a special topics course which the department would offer as an elective course in the future; the department aims to offer one elective per semester A. Proposal significance and objectives. Once installed, the instrument would be immediately used in the following (representative) research activities of the department: Isolation and Analysis of Proteins (Dr. Kelly Gallagher) HPLC was introduced into the biochemistry lab curricula of most programs over 15 years ago (based on a review of chemical education literature). HPLC has never been employed in our biochemistry laboratory in spite of the fact that its use in modern research has continued to grow and evolve. Because HPLC has been so useful in the study of biomolecules, the American Chemical Society, the professional organization that certifies our program, stated, it is essential that future chemists know how to use this technique routinely (HPLC: A Practical Technique for Future Chemists, J. Chem. Educ., 1992, 69 (4), p 260). HPLC would enhance work on a collaborative project with Nancy Bachman of the Biology Department. Several undergraduates are currently attempting to determine the structures, oligomeric states, and binding properties of the newly discovered proteins CGI-112, COX4N, and TTC35 whose behavior appear to play a role in various diseases including Parkinson s disease and cystic fibrosis. The availability of an HPLC would enable us to conduct these studies on campus, eliminating the need to travel and purchase equipment time at other institutions. Bioogranic Chemistry (Dr. Maurice Odago) Chirality is a feature of many molecules and a pair of chiral molecules are identical except in the spatial orientation of their atoms. The separation of chiral compounds continues to be of great interest because the majority of bioorganic and organic molecules are chiral. In addition, one the areas of increasing interest in organic synthesis is homogeneous chiral catalysis. As with any chemical synthesis, purification and characterization (identification) is necessary, and one of the ways of achieving both is with the use of chiral stationary phases in HPLC. The availability of the proposed HPLC will provide my research students with a quick turn-around times for analysis and characterization while saving the student a lot of time spent on resolution (separation) techniques. This is especially important in in an undergraduate research environment where students often do not have large blocks of time within which to do experiments. Analytical and Environmental Chemistry (Dr. Trudy Thomas-Smith and Dr. Schaumloffel) Current interest in the analysis of water samples for the presence of active ingredients in pharmaceuticals and personal care products (PCPPs) continues to increase as agencies across the country are charged with ensuring safe public waters supplies and natural waterways. Investigation of the presence and fate of the PCPPs in various water samples (including natural waters) is currently being pursued by this investigator primarily using diode array spectroscopy. However this technique on its own does not offer
3 Department of Chemistry and Biochemistry StAR Proposal the sensitivity required for the detection of small amounts of a compound and so separation and/or concentration of compounds of interest is necessary. The use of HPLC/DAD will minimize or eliminate the need for the labor-intensive sample preparation requirements and will allow for the structural elucidation of the degradation products anthropogenic compounds in the natural water environment, this work would be complementary to a growing body of research involving so-called emerging contaminants. In light of the above-mentioned research projects therefore, We believe such equipment will enable the students to juggle with many activities of their daily courses, while still given then an opportunity to gain optimal experiential learning and better prepare them for future careers in research, jobs and higher education in chemistry. B. Contribution to the SP2010. The instrument will be used directly in faculty-led undergraduate student research. Undergraduate research experience has been shown to valuable both in terms of student understanding and graduate marketability. Participation in the research projects facilitated by the proposed HPLC is well-aligned with the College s academic master plan recently approved by the college senate, which places students at the center of teaching learning and scholarship and is central to the 2010 strategic plan ( ). C. Priorities Addressed We strongly believe that the acquisition of an HPLC-DAD aligns well with all the priorities listed by the reviewers, although the argument can be made that academic quality and rigor and student engagement are the priorities best epitomized by the use of this instrument in undergraduate research and in teaching. a. Academic quality & rigor An important aspect of the undergraduate chemistry student s curriculum is exposure to and the study of chromatography which is a separation technique and HPLC is the most widely used chromatographic method. Comprehensive understanding of HPLC requires an understanding of concepts learned in General (Introductory) and Organic Chemistry, the foundational courses for our curriculum. Furthermore, the premier professional organization of chemists in the United States, the American Chemical Society (ACS) which offers a degree approval program (our department is ACS-approved) lists chromatography as a topic that is required to be covered in the bachelor s level program in two of the five chemistry sub-disciplines: Analytical Chemistry and Biochemistry. b. Experiential learning opportunities for students Experiential learning is critical to the study of chemistry, and much of the learning and understanding of chemistry takes place in the laboratory setting. Further, hands-on experience with instrumental methods have proven time and again to be useful to students in their future endeavors as scientists in various settings. In fact, a survey of potential employers of chemistry graduates suggests that students are not necessarily gaining the hands-on instrumental experience desired by employers and more than 66 % of industrial employers list experience with HPLC as a desired skill for chemistry graduates. c. External revenue generation Our department has been active and successful in seeking external funding for scientific instrumentation. More recently however, increased competition, has resulted in recommended proposals not being funded. One area that could strengthen our case for external funding is the inclusion of preliminary data in our proposals. The research projects listed all have the potential for growth into larger and externally collaborative projects and given the point at which the proposing faculty members are in their career trajectory, application for external funding is a certainty.
4 Department of Chemistry and Biochemistry StAR Proposal d. Faculty growth, success, and retention It is (arguably), difficult to separate academic quality and rigor from faculty success. Certainly if our goal in the strategic and academic master plans are to be realized, student access to a high quality program is linked to and may be viewed as a measure of faculty success. Further, the college s ability to provide direct support for the pursuit of experimental science must be viewed as a predictor of (science) faculty satisfaction and therefore retention. e. Quality of undergraduate experience Graduates of the chemistry program have stated that hands-on experience in the use of instrumentation has served them well in their vocations after graduation. Indeed D. Impact on operational efficiency and effectiveness. The proposed acquisition directly impacts the research activities of three tenure/tenure-track faculty members each in distinct sub-disciplines of chemistry. Therefore, the HPLC-DAD should not be viewed as a highly specialized tool. In addition to these already identified areas of use the HPLC-DAD can be potentially be used in Organic and Physical Chemistry courses. E. Opportunity for Collaboration. Since the primary use of an HPLC is the separation and identification of compounds, there are therefore opportunities for collaboration with many different academic areas. Case in point: faculty members from the departments of Biology and Human Ecology have approached us seeking assistance with compound identification and HPLC/DAD is well-suited to this purpose. It should also be noted that Dr. Gallagher s work involves collaboration with Dr. Nancy Bachman and Dr. Odago works closely with Dr. Bill Vining (inorganic chemist) in our department. F. Effect on Department Goals The implementation of experiments using the proposed HPLC/DAD will directly impact five of the ten department learning objectives including collaborative investigation of scientific knowledge, competence in analytical thinking and effective communication skills. Research activities executed using this instrument will facilitate student presentation at national meetings and may result in publication in peer reviewed journals each of which are specifically listed in our department s goals in our program review. G. Assessment Direct assessment in the context of student learning outcomes will take place. For example student learning outcomes in Biochemistry are assessed via a variety of assignments developed from material in recent research literature. The majority of student research culminates in presentation at a formal conference or meeting. It is further reasonable to anticipate that pedagogical and/or research-based articles will be published based on data generated with this instrument H. Implementation strategy and timeline. Instrument installation and initial training will be provided upon purchase and the undergraduate research projects described will begin as soon as is practicable (Fall 2013). The proposed elective course will be designed and tested in the summer of Routine preventative maintenance of the instrument is not significantly time consuming, and two faculty members (Drs. Schaumloffel and Thomas-Smith) are experienced in the use and maintenance of HPLC instrumentation. Finally some consumables are already available in the department and others will be supplied by the regular department budget.
