Structure 6 Regional Official Laboratories 2

Tasks Main areas of activity I. Food microbiological diagnostic activity official control laboratory for the capital Budapest and county Pest, investigation of foodborne disease associated samples - all the country specific tests all the country (eg. VTEC, Cronobacter, detection of bacterial toxins, GMO, species identification, virus detection) EU surveys Salmonella, Campylobacter, Listeria testing for export requirements (eg. USA export) II. Monitoring activity organisation of the microbiological monitoring sampling system microbiological testing evaluation and reporting the results 3

Tasks Main areas of activity III. National Reference Laboratory Tasks Salmonella Listeria monocytogenes Campylobacter E. coli Microbiological contamination of milk and milk products Coagulase positive Staphylococci GMO II. Central role in the laboratory system organisation of ringtests microbiological working groups, training 4

Methods NRL Salmonella Accreditation according to since 1995 ISO 6579 o Detection: MSZ EN ISO 6579:2006/A1:2007 D will be changed to the new standard method /year o Serotyping: MSZ CEN ISO/TR :2014 o /year Phage typing: S. Enteritidis and S. Typhimurium, Colindale method o ~ 200 /year Vaccine strain differentiation IDT vaccines Salmovac SE: minimal medium inoculation and Kylt PCR IDT vaccines Zoosaloral H: minimal medium inoculation Avipro (TAD) Salmonella Vac E and Vac T: antimicrobial resistance by Avipro Plate 5

Methods NRL Salmonella Molecular detection Real-Time PCR: Salmonella Enteritidis and Typhimurium detection Foodproof S. Enteritidis and Typhimurium Kit 2013 Real-Time PCR: Bio-Rad iq-check Salmonella II. PCR Detection Kit Monophasic S. Typhimurium confirmation (Tennant et al., not accredited) Kylt SE DIVA 1. (not accredited) 6

Methods NRL Salmonella Molecular typing PFGE Other laboratory PT strains, foodborne outbreaks NGS, WGS Plan: to build up a high capacity NGS laboratory that serves all the NGS needs from different disciplines (food microbiology, veterinary diagnostics, plant health, identification of breeding animal etc.) Until that: we have just initiated a contract with a commercial lab providing NGS services (~ 280 EURO/ 1 strain, WGS with bioinformatical analyses of the data, therefore it will be used only for outbreak investigation) 7

Coordination NRL Salmonella Coordination the activities of official laboratories (6) All the official laboratories are accredited according to ISO and ISO 6579 Organisation of proficiency tests Salmonella detection Participants: official and private labs Matrix: food, poultry faeces Positive samples: culture method Exclusion of S. Enteritidis and S. Typhimurium by O-serogrouping (the method is used at broiler flocks before slaughter) Participants: official and private labs Matrix: Salmonella isolates 8

Coordination NRL Salmonella Coordination the activities of official laboratories (6) Training of new colleagues joining to the regional labs, new methods Training of colleagues to new methods Organize laboratory meetings regularly Information and news concerning the laboratory system Provide information and technical assistance for the implementation of monitoring plans and other specific duties Dissemination of information about sampling and analysis methods Discussion of emerging problems Discussion of scientific or methodological subjects ISO 6579:2017 9

Collaboration NRL Salmonella Collaboration with the EURL-Salmonella Participation on the yearly workshops Participation in proficiency tests Discussion of methodological problems Outbreak investigation Foodborne outbreak related to eggs in Europe Isolates from Hungary 12 allels difference 10
