3 . Eclectica is a bench top analyzer consisting of two carousels, one to process samples and the other to process reagents, each with its own barcode reader. Technical Characteristics. Small-in size (85 cm. wide, 66 cm. deep, and 54 cm height).. Eclectica fits easily on any lab bench.. No hydraulic or wall connections are required.. The touch screen, color-integrated monitor, together with the separate, alphanumeric keyboard, provides the user with a friendly and fast interface.. An embedded PC controls all steps from primary tube allocation to the printing results.. Download and upload data through a direct connection between Eclectica and a Laboratory Information System (LIS). With its user-friendly software Open-Lab and immediate graphs, Eclectica, designed for simplicity, checks analytical and technical performance in real time. 4 ECLECTICA TEST MENU 1. Arm A double arm with two magnetic needles, one for samples and calibrators, the other for specific reagents, to avoid cross contamination Smart card reader A smart card reader, located on the front side of the instrument, reads the smart card included in the reagent packaging and loads data related to specific reagents, calibrators, common reagents. 3. Printer An internal, thermal printer is located on the top right side of the device, prints the results. 5 Technical Features - Operational 4. Reagent carousel Reagent position on the analyzer is transmitted by a self-turning, barcode reader. The instrument uses levels and an automatic counter for double verification. The time for work list preparation is short and risk of reagent misclassification is avoided. Ready-to-use reagent vials can be randomly allocated. There are 20 positions defined for specific reagents and 28 positions defined for calibrators. Because the reagent carousel is cooled, reagents can remain on board for several days. The analyzer is always ready, saving space in the laboratory refrigerator and simplifying the daily routine 5. Samples carousel The carousel has sixty positions reserved for primary test tubes, ranging in size from 12/13 mm. by 50/100 mm. Test tubes can be loaded directly on board for Immunoassay and Clinical Chemistry tests. Disposable, self-emptying reaction well segments reduce the risk of contamination and time spent in «ready mode». There are four segments and each segment has 24 positions for an overall total of 96 positions. In the middle of the carousel are diluents for Immunoassay tests as well as a magnetic, solid phase container. The unit is compact without outside containers. The Substrate, Stop solution, Wash solution and liquid waste tanks are all located on the right side of the unit. 6. Monitor A 12-inch, flat, touch screen video accepts input from touch or pen. 6 Immunoassay and Clinical Chemistry: Method Principles a General Eclectica employs a simple, proven, immuno-reaction scheme (IEMA - EIA) based on magnetic particle immunoassay. The separation method is versatile and can be applied to many molecule types, from small molecules to larger molecules, using the same separation reagents in different assay. The high surface area of magnetic particles allows for high sensitivity and a wide range of linearity.. Direct Immuno-enzymatic Assay (IEMA). Competitive Immuno-enzymatic Assay (EIA) a Immunoassay: Method Principles The Eclectica reaction scheme for Immunoassay includes reagents for all reaction steps:. Monoclonal/Polyclonal antibodies for immunoreaction.. Separation reagents consisting of magnetic particles linked to an antibody specifically for FITC for magnetic solid phase separation reaction.. Enzyme/Substrate for color development.. All the reagents are ready to use.. Minimum reaction volumes assure rapid testing and reduction of liquid waste. a Direct Immuno-enzymatic Assay (IEMA) In the Immuno-reaction for the direct immunoassay, the sample incubates with two carefully selected antibodies. These antibodies react to the subject analyze by forming a sandwich. 7 a Competitive Immuno-enzymatic Assay (EIA) In the immuno-reaction for the competitive EIA, only one alkaline phosphates labeled antibody is used. A FITC conjugate reagent is added that competes with the sample analyte for the limited number of binding sites available on the small amount of added antibody. a Magnetic Particles After the first incubation, magnetic particles («Separation reagent») are added that react with the FITC-labeled antibody to form an immuno-complex. The application of a magnetic field allows for B/F separation. a Colour Development Enzymatic degradation of the substrate by the alkaline phosphatase, through endpoint kinetics, gives an analytical signal. The flexibility of this method makes possible testing of both small (such as Haptens) and large molecules using a universal separation reagent, substrate, stop solution and washing solution. Simultaneous reading at two wavelengths means accurate readings on a wide concentration of ranges. a Clinical Chemistry The Clinical Chemistry assays are according to IFCC recommendation and German standardization (DGKC). 9 In every IA method, a master curve is read directly from the smart card and stored on board. The software uses tcontrols to check the test validity. Positive barcode ID of samples and reagents, master curve calibration, refrigerated on-board reagent carousel, internal and external washing needle all combine to guarantee a complete and reliable system.
