4 Priming and Calibration of Diatron Analyzers REAGENT CONSUMPTION in ml Diatron Abacus Priming of Diatron Abacus 1. Take the aspirating tubes from the reagent cubitainers of the original reagents. Don t lay the tubes on tissues or dusty surfaces. Place them in clean beakers. 2. It is important to rinse the outside surface of the aspirating tubes with distilled or de-ionised water to be aware of any kind of reagent contamination. 3. Then prime the diluent. Select menu 3: Special/ Maintenance. Press OK. Select menu 1: Maintenance. Press OK. Select menu 2: Prime. Select Prime diluent. The first priming is intended to aspirate some air between the original and Avantor Performance Materials Diluid Abacus. Place the aspirating tube of the diluent after the first priming into the cubitainer of Diluid Abacus. Perform now another 3 priming cycles to be sure that Diluid III Diff is completely primed. 4. After priming the diluent, prime the lyse. Perform the same steps as priming diluent but select at the last step Prime lyse. The first priming is intended to aspirate some air between the original and Avantor CyMet Abacus. Place the aspirating tube of the lyse after the first priming into the cubitainer of CyMet Abacus. Perform now another 3 priming cycles to be sure that CyMet Abacus is completely primed. 5. The last reagent to be primed is the cleaning solution. Perform the same steps as priming diluent but select at the last step Prime cleaner. The first priming is intended to aspirate some air between the original and Avantor Proclean Diatron Abacus. Place the aspirating tube of the cleaner after the first priming into the cubitainer of Proclean Diatron Abacus. Perform now another 3 priming cycles to be sure that Proclean Diatron Abacus is completely primed. 6. All reagents can be primed in one cycle. Select at the last Prime all. Use 8-, or 12 parameter Hematology blood control for automatic calibration Caution: New calibration will invalidate the previous one, which can never again be retrieved. 1. In factorial mode the calibration measurement must be performed in normal measuring mode and after the calculating, the user must set the calibration factors manually in the CALIBRATION menu. Select menu 6: Settings. Press OK. Select menu 5: Calibration settings. Press OK. Select menu 1: Calibration mode. Press OK. Select Factorial. Go back to the Main menu. Select menu 4: Calibration. Press OK. Select menu 1: Calibration. Pressing numeric keys and OK to enter can do factor settings. Factors must be between 0.8 and In automatic mode, after settings of the reference values (use Coulter assay values), the analyzer performs calibration measurements and calculates the new calibration factors that could be validate by the confirmation of the operator. 5 Select menu 5: Calibration settings. Press OK. Select menu 1: Calibration mode. Press OK. Select three measures. Go back to the Main menu. Select menu 4: Calibration. Press OK. Select menu 1: Calibration. Press OK. After setting target values, measure hematology blood control (8 parameter or 12 parameter control) three times by pressing the sampling bar. First the target values must be defined by the numeric keys and OK to enter. Use Coulter assay values. If you do not want to calibrate a parameter, leave its target value 0. Press to go to calibration measurement. Values of the calibrated parameters can be set in the following ranges: Parameter Low limit High limit WBC HGB g/l RBC HCT MCV RDW CV PLT MPV 5 15 PCT 0 2 PDW CV 5 50 Press to accept results. The actual and the total number of calibration measurements are shown in the first line. After calibration the new factors are displayed. 6 introduction Diatron Abacus Junior 5 Priming of Diatron Abacus Junior 5 All reagents can be primed from the Priming Menu. From the Utilities Menu press <1. Maintenance>. Then press <2. Prime all Calibration of Diatron Abacus Junior 5 There are two ways of calibration: factorial or automatic. Use 3-diff or XE-diff Hematology blood controls for automatic calibration (Be aware that 3-diff doesn t give you values for the EOS and BASO). Please refer to the User s Manual of the Abacus Junior 5 for calibration details. 1. Note: Use 5-part diff reference machine or other reference method to calibrate Eosinophils and Basophils. 2. In automatic mode, after settings of the reference values (use XE-diff assay values), the analyzer performs calibration measurements and calculates the new calibration factors that could be validate by the confirmation of the operator. From the Calibration menu press <3: Calibration Settings>. Select 4 or 5: Calibrate with one or three measurements. Go back to the calibration menu. Select 1: Calibration. First the target values must be defined by the numeric keys and OK to enter. Use XE-diff assay values. If you do not want to calibrate a parameter, leave its target value 0.
