E - m ail: huangxienan@ yahoo. com. cn) [ ABSTRACT] A IM : To study the role of p rostaglandin F 2 ( PGF 2 ) in cardiac hypertrophy and its relation with calcineurin (CaN) signal transduction pathway in vitro. M ETHOD S: The cultured neonatal rat cardiomyocyte was used to observe the hypertrophic effect of PGF 2, and the hypertrophic response was assayed by measuring the cell diameter, p ro2 tein content and atrial natriuretic factor (ANF) mrna exp ression. For mechanism studies, the intracellular free calcium concentration ( [ Ca 2 + ] i ) in cultured cardiomyocytes was measured by using Fura - 2 /AM as a fluorescent indicator. ANF and CaN mrna exp ressions, and the exp ressions of CaN and its downstream effectors, NFAT 3 and GATA 4 p roteins were assayed by RT - PCR and W estern blotting, respectively. RESUL TS: In cultured cardiomyocytes, PGF 2 induced p rofound hypertrophic morphology change and the significant increase in cell diameter, and p rotein content in a concentration - de2 pendent manner compared with those in vehicle control ( P < 0101). The same result was found in measuring the [ Ca 2 + ] i in cardiomyocytes ( P < 0101). PGF 2 at concentration of 10-7 mol/l significantly p romoted ANF and CaN mrna exp res2 sions and the p rotein exp ressions of CaN /NFAT 3 /GATA 4 compared with those in the vehicle control ( P < 0105). Cyclos2 porin A, a CaN inhibitor, markedly inhibited the myocyte hypertrophy ( P < 0101), reduced the increased [ Ca 2 + ] i ( P < 0101), and decreased the exp ressions of CaN mrna and CaN /NFAT 3 /GATA 4 p roteins ( P < 0105) compared with those of only PGF mol/l treatment.
