1 Grade: 9 th Subject: Biology Author: Ladys Contreras Unit: Plant Structures Content Standard (TEK 5B/SEs): The student knows how an organism grows and the importance of cell differentiation. The student is expected to examine specialized cell, including roots, stems, and leaves of plants; and animal cell such as blood, muscle, and epithelium. Content Objective: Student will be able to examine the vascular tissues (xylem and phloem) of the stems and roots in plants. Key Vocabulary: Vascular tissues, xylem, phloem, ground tissue, lignin, cell wall, cellulose, toluidine blue Language Objective ELPS 74.4 (5B): The student is expected to write a paragraph explaining their observations of the vascular tissues under the microscope using accurate spelling and the key vocabulary words. Materials: Activity sheet, small cross-section of the stem and root, water, slide, Toluidine blue, coverslip, microscope, forceps, pipette, tube, gloves, lab coat, agree and disagree cards, switchgrass Learning Activities: Engage: 1. Teacher will review the content and language objectives with the students. 2. The teacher will ask the students, What do you know about vascular tissues? Why are vascular tissues important to plants? The rest of the students will show a card to indicate whether they agree or disagree with the answers. Explore: 1. Teacher guided review of vocabulary key words using the Frayer model (graphic organizers that requires students to define target vocabulary and apply their knowledge by generating examples and non-examples, giving characteristics, and/or drawing a picture to illustrate the meaning of the word.) 2. Teacher will review with students the lab protocol about staining vascular tissue of switchgrass. 3. Teacher will model the experiment to the students in the lab. 4. Students will work in pairs to carry out the experiment using the protocol. Explain: 1. Student will work in pairs to draw and label the observations of the tissues seen under the microscope. 2. Student will demonstrate their understanding of vascular tissues completing the lab report. Evaluate: 1. Student will write a lab report. 2. Student will complete a crossword activity about the key vocabulary words of vascular tissues in plants. Adaptations for ELs: Beginner: The student will write a caption for a picture that illustrates their observations under the microscope with the new vocabulary words. Pre-teach vocabulary words. Intermediate and Advanced: The students write a brief paragraph explaining their observations under the microscope using complete sentences and appropriate vocabulary.
2 Lab Report: Vascular tissues Materials Transverse section of the stem and Forceps Pipette root Toluidine Blue stain Water Slide Coverslip Microscope Lab coat Tube Gloves Procedure: 1. Add the blue colored dye (Toluidine Blue) into the tube with the small pieces of stem. 2. Wait for one minute. 3. Remove the excess of the stain with the pipette. X 3 6. Repeat three times. 4. Add one full pipette of water in the tube. 5. Remove the excess of water with the pipette.

Be sure to label each drawing with the type of material it represents. Write a brief paragraph explaining your observations under the microscope using complete sentences and appropriate vocabulary.
