Abstract of thesis presented to the Senate of Universiti Malaysia Terengganu in fulfillment of the requirement for the degree of Master of Science MORPHOLOGICAL, GENETIC AND BIOLOGICAL STUDIES OF RED PALM WEEVIL, Rhynchophorus spp. (COLEOPTERA: CURCULIONIDAE) IN August 2016 Main supervisor Co-Supervisor School : Associate Professor Wahizatul Afzan Azmi, Ph.D. : Chong Ju Lian, Ph.D. : School of Marine and Environmental Sciences The Red Palm Weevil (RPW), Rhynchophorus ferrugineus (Olivier) is the most important pest of various palm species including coconut, date and oil palm in the world. Rhynchophorus ferrugineus is a lethal pest of coconut in Terengganu and sago palm in Sarawak, Malaysia. This study was intended to investigate the morphological, genetic and biological studies of red palm weevil (RPW) which include of two Rhynchophorus morphospecies R. ferrugineus and R. vulneratus in Malaysia. For morphological aspect, body length dan rostrum length of R. ferrugineus female were significantly longer than males (p < 0.05). However, the elytra length, lengths of foreleg, mid leg, hind leg and antennae length of R. vulneratus males were significantly longer than females. Overall, all measurement of morphological characters R. vulneratus males and females were significantly larger than R. ferrugineus adults (p < 0.05). Scanning Electron Microscope (SEM) analysis showed no differences on certain microstructures between two Rhynchophorus morphospecies. For genetic aspect, mitochondrial DNA cytochrome c oxidase subunit 1 (COI) were used to investigate genetic variation among 11 selected morphs of R. ferrugineus in order to determine if ii

they were of the same species and if whether they are genetically identical. The results show that 11 different morphs of R. ferrugineus were genetically similar, and were 100% identical to each other. The highly dissimilarity percentages of nucleotides bases were found between R. ferrugineus and R. vulneratus for COI gene. Thus, it could be concluded that R. ferrugineus and R. vulneratus are different species sharing the same genus. The reproductive performance of R. ferrugineus were studied using 9 pairs of virgin adult weevils under laboratory conditions. The overall life cycle of R. ferrugineus reared on sago diet was about five months. The number of fecundity until survived larvae (until 7th instar) took approximately 81 days and were decreased gradually for each pair of R. ferrugineus. The adult of R. ferrugineus were successfully emerged from cocoon. In conclusion, the measurement of morphological characteristics and molecular genetic analysis suggested that R. ferrugineus and R. vulneratus are distinct species. The life cycle of R. ferrugineus took around five months as compared to previous studies. It is hoped that the findings from this study will provide important information on the morphological, genetic and biological aspects of this invasive pest, specifically in tropical climate of Malaysia. iii

Abstrak tesis yang dikemukakan kepada Senat Universiti Malaysia Terengganu sebagai memenuhi keperluan untuk ijazah Sarjana Sains KAJIAN MORFOLOGI, GENETIK DAN BIOLOGI KUMBANG PALMA MERAH, Rhynchophorus spp. (COLEOPTERA: CURCULIONIDAE) DI Ogos 2016 Penyelia Utama Penyelia Bersama Pusat Pengajian : Profesor Madya Wahizatul Afzan Azmi, Ph.D. : Chong Ju Lian, Ph.D. : Pusat Pengajian Sains Marin dan Sekitaran Kumbang palma merah (RPW), Rhynchophorus ferrugineus (Olivier) adalah kumbang perosak utama untuk pelbagai spesies palma termasuk kelapa, kurma dan kelapa sawit di dunia. Rhynchophorus ferrugineus adalah perosak kelapa di Terengganu dan sagu di Sarawak, Malaysia. Kajian ini bertujuan untuk mengkaji morfologi, genetik dan biologi kumbang palma merah (RPW) iaitu termasuk dua morfospesies Rhynchophorus R. ferrugineus dan R. vulneratus di Malaysia. Untuk aspek morfologi, panjang badan dan panjang rostrum R. ferrugineus betina adalah lebih panjang secara signifikan berbanding dengan kumbang jantan (p < 0.05). Walau bagaimanapun, panjang elytra, panjang kaki depan, kaki tengah, kaki belakang dan panjang antennae R. vulneratus jantan adalah lebih panjang secara signifikan berbanding dengan kumbang betina. Secara keseluruhannya, semua pengukuran ciri morfologi kumbang jantan dan betina R. vulneratus adalah signifikan lebih besar daripada kumbang dewasa R. ferrugineus (p < 0.05). Analisis SEM menunjukkan tiada perbezaan untuk sesetengah mikrostruktur antara dua morfospesis Rhynchophorus. Untuk aspek genetik, mitokondria DNA cytochrome c oxidase subunit 1 (COI) telah iv

digunakan untuk mengkajikan variasi genetik antara 11 morfo terpilih daripada R. ferrugineus untuk mengesahkan sama ada ianya adalah spesies yang sama dan mempunyai genetik yang serupa atau tidak. Keputusan menunjukkan bahawa 11 morfo R. ferrugineus mempunyai genetik yang sama, dan 100% adalah sama antara satu sama lain. Terdapat perbezaan yang ketara dalam nukleotida antara R. ferrugineus dan R. vulneratus untuk gen COI. Oleh itu, ia boleh disimpulkan bahawa R. ferrugineus dan R. vulneratus adalah spesies yang berbeza yang berkongsi genus yang sama. Prestasi pembiakan R. ferrugineus telah dikaji dengan menggunakan 9 pasangan kumbang dewasa dara di bawah keadaan makmal. Keseluruhan kitaran hidup R. ferrugineus yang dibela dengan menggunakan diet sagu adalah kira-kira lima bulan. Tempoh kesuburan sehingga larva yang hidup (sehingga instar 7) mengambil masa kira-kira 81 hari dan telah menurun secara beransur-ansur untuk setiap pasangan R. ferrugineus. Dewasa R. ferrugineus telah berjaya keluar daripada kepompong. Secara keseluruhannya, pengukuran ciri-ciri morfologi and analisis genetik molekul mencadangkan R. ferrugineus dan R. vulneratus adalah spesies yang berbeza. Kitaran hidup R. ferrugineus mengambil masa lebih kurang lima bulan berbanding dengan kajian terdahulu. Penemuan dari kajian ini diharapkan dapat memberikan maklumat penting dari segi aspek morfologi, genetik dan biologi kumbang ini, khususnya dalam iklim tropika Malaysia. v
