1. Materials. Unless otherwise stated, reagents and solvents were commercially obtained and use without further purification. 2. Measurements. The absorption spectra were measured on a Hitachi U-3010 spectrometer. Fluorescence spectra were measured on a Hitachi F-4500 fluorometer. 1 H NMR and 13 C NMR spectra were obtained on a Bruker Avance DPS-400 spectrometer, respectively. MALDI-TOF mass spectrometric measurements were performed on Bruker Biflex III MALDI-TOF (both positive and negative ion reflector mode). 3. Methods. 3.1 Determination of the binding constant The binding constant K of MCy-1/Hg 2+ complex were determined from the following Benesi-Hildebrand equation 12 : Where K = Binding constant; A 0 = The observed absorption in the absence of cation; A = The observed absorption the cation-added; [M] = The concentration of the cation-added; a and b are constants, the binding constant value K was evaluated graphically by plotting 1/A against 1/[M] 2

3.2 Determination of the detection limit The detection limit DL of MCy-1 for Hg 2+ was determined from the following equation: DL = K*Sb1/S Where K = 2 or 3; Sb1 is the standard deviation of the blank solution; S is the slope of the calibration curve. 4.Photophysical Properties of MCy-1 and MCy-1/Hg 2+. 4.1 Photophysical Properties of MCy-1 and MCy-1/Hg 2+ compounds ab ( nm ) em ( nm ) a f b MCy-1 695 777 86 000 0.101 MCy-1/ Hg 2+ 817 777 190 000 0.028 (a) Molar extinction coefficients are in M -1 cm -1 and in the maximum of the highest peak. 6. Synthesis Compound 1 and 2 ( dithia-dioxa-monoaza crown ether) were synthesized according to the literature. 1, 2 Synthesis and characterization data of MCy-1 Preparation of MCy-1. 79.5 mg (0.1 mmol) of cyanine and 125 mg (0.5 mmol) of dithia-dioxa-monoaza crown ether were dissolved in 10 ml anhydrous DMF in a 25mL round bottom flask. The mixture was stirred at 90 C under argon for 3 hours. Then it was cooled to room temperature and added to 120 ml of ether with violent stirring. The obtained blue solid was filtered and dried under vacuum, then purified on column chromatography on silica gel using methanol-dcm mixture as eluent to give an dark blue product. (53.5 mg, 53%). 1 H NMR (400 MHz, DMSO-d 6 ): 1.23 (m, 2H, CH 2 ), 1.68 (s, 12H), 2.28-2.76 (t, 4H, J = 4.88 Hz), 2.96 (t, 4H), 3.74 (t, 4H, J = 4.88 Hz), 3.83 (s, 4H), 5.34 (s, 4H), 5.97-6.00 (d, 2H J = 13.4 Hz, CH), 7.15-7.17 (d, 2H, CH), 7.23-7.25 (d, 4H, CH), 7.28-7.30 (d, 2H, CH), 7.34-7.38 (m, 4H, CH), 7.55-7.58 (m, 4H, CH), 7.74 (s, 2H, CH). 13 C NMR (150 MHz, DMSO-d 6 ): 21.8, 24.5, 29.1, 29.2, 29.6, 31.3, 32.2, 32.5, 36.3, 40.6, 46.9, 48.3, 56.2, 70.9, 73.5, 98.1, 109.6, 120.4, 125.3, 126.6, 127.1, 128.1, 129.5, 136.0, 140.1, 142.3, 143.5, 143.6, 144.8, 162.9, 169.9, 173.5.
