High temperature catalytic combustion with Infrared Detection (NDIR) is used for the TC analysis and the determination of TN is based upon the well-proven DUMAS methodology using high temperature combustion and detection with Thermal Conductivity (TCD). Principle of operation The samples are weighed in a quartz crucible and placed in the autosampler. By a unique vertical bottom-to-top sample introduction system the samples are introduced in the high temperature combustion furnace. At 1100 C the Carbon is completely oxidized to CO 2 and Nitrogen is converted into NxOy in the presence of Oxygen. After the catalyst furnace, where the oxidation reaction takes place, the sample passes through a Peltier cooler to remove moisture formed by condensation. Then the sample gas is splitted and transferred, by the carrier gas Helium, to the reduction oven. In the reduction oven of 600 C NxOy is reduced, in the presence of a copper reductor, to N 2. Then first the CO 2 is measured by Non Dispersive Infra Red Detection (NDIR) for Total Carbon and subsequently the by-products, water and carbon dioxide are removed by an absorber. Secondly the N 2 gas is measured by Thermal Conductivity Detection (TCD) and to remove interferences a background correction is performed by measuring the He gas.
Data Acquisition and Instrument Control The Primacs SNC analyzer is controlled by Skalar s flexible SN-Access data acquisition software. The software is easy to set up and very user-friendly. Using pull-down menus for different actions it allows the operator to start analysis within a few minutes. During analysis the worklist can be modified, to enable the addition of priority samples, samples or sample positions can be edited or deleted. A dual display is available during the run to monitor both the Carbon and the Nitrogen channels in real-time simultaneously. In addition other information such as temperatures and detector signals, which are of major importance for reliable elemental analysis, are available in real-time. All analytical results and calibration curves can be viewed and edited in the results window. The results can easily be printed or exported to a LIMS system. DUMAS versus Kjeldahl technique The Skalar Primacs SNC uses the automated DUMAS combustion technology for measurement of the total nitrogen/ protein concentration. In the Dumas method, a sample is burned in an oxygen rich atmosphere at high temperature and the resulting gases are analyzed. The Dumas combustion method has several advantages over the Kjeldahl technique in terms of speed and its environmental acceptability. In addition the Dumas method does not require any additional sample pretreatment such as digestion with toxic chemicals. The large sample size capacity of this instrument ensures reduced sample handling time and optimal automation. Detection is based on thermal conductivity (TCD). TCD detectors are able to process an extensive working range to cover nitrogen/protein levels from low parts per million to high percentage concentrations. The DUMAS combustion technology offers an accurate, rapid, safe, economical and environmentally friendly method for Nitrogen / Protein determination.
