1 25-OH VITAMIN D 3 IN PLASMA BY UV FAST CODE Z19610 BIOCHEMISTRY Quantification of 25-hydroxyvitamin D 3 (25-hydroxycholecalciferol) in serum is the best established indicator of vitamin D status. Vitamin D 3 (cholecalciferol) is absorbed with the diet and, given sufficient ultraviolet irradiation, nutritionally adeguate amounts of vitamin D 3 are formed in the skin from its precursor, 7-dehydrocholesterol. In the liver, vitamin D 3 undergoes hydroxylation to 25- hydroxyvitamin D 3, which is further metabolized in the kidney to form the active metabolites 1,25- hydroxyvitamin D 3 and subsequently 24,25-hydroxyvitamin D 3. The prevalence of hypovitaminosis D has been recognized as substantial, even in regions with high sun exposure, contributing not only to osteoporosis but possibly to a loss of muscle strength in aging as well. 2 TECHNICAL FEATURES Principle of the Method 25-OH Vitamin D 3, after deproteinization with a specific reagent, and after purification with Clean-up Columns is injected directly into isocratic HPLC system. Recovery : > 93% Sensitivity : about 2 µg/l Linearity : µg/l Normal Range : µg/l Components of the kit : All the components are ready-to-use and stable 3 years at 2-8 C. Pre-analytical Step : Plasma samples should be stored at 20 C for no longer than 3 months. 2

3 WARNING: USE ONLY GLASS TUBES FOR EACH OPERATIVE STEP! DON T use absolutely plastic tubes! PREANALYTICAL PROCEDURE Pipette in a glass tube : 400 µl of Mobile Phase 100 µl of Reagent F - Test Solution INJECTION : Vortex for 10 seconds Inject 100 µl of this solution in the chromatographic system. Verify that the Vitamin 25-OH- D 3 has retention time similar at fig. 1. If the Test is all right you can start with the analytical procedure; if not, check the functionality of the analytical system. Important : Don t use this solution to calibrate! ANALYTICAL PROCEDURE STEP 1 : Calibrator, Controls and Samples Preparation: Pipette in a glass tube: Calibrator Control Sample Calibrator 1,0 ml Control 1,0 ml Sample 1,0 ml Reagent A Deproteinization Sol. 1,5 ml 1,5 ml 1,5 ml Warning: deproteinize directly on Vortex for at least 10 sec. STEP 2 : Centrifuge at rpm for 5 min. STEP 3 : Place the surnatant (about 2,0 ml) in a clean glass tube. Add 0,5 ml of H 2 O HPLC grade. STEP 4 : Clean-up Column Attivation: Vortex for 10 seconds 1,0 ml of Reagent B Conditioning Sol. N 1 and discharge the percolated liquid 1,0 ml of Reagent C Conditioning Sol. N 2 and discharge the percolated liquid STEP 5 : Samples, Calibrator and Controls loading into Clean-up Columns: Dispense the surnatant (about 2,5 ml), previously prepared, into clean up columns. (As Clean-up Columns is 1,0 ml, pipette 1 ml of surnatant each time). Discharge the percolated liquid, if needed a vacuum system could be used for percolation, at speed of 1 ml / min ( drop by drop ). STEP 6 : Clean-up Column Wash: Pipette in Clean-up Columns 1,0 ml of Reagent D Wash Solution and discharge the percolated liquid. Release N 006 Vitamin 25-OH-D3 in plasma by UV - FAST August

4 STEP 7 : Elution of 25-OH-D 3 : Pipette in Clean-up Columns 350 µl of Reagent E Eluting Sol. Collect the eluate in a glass tube Add 50µl of H 2 O HPLC grade Vortex for 10 seconds N.B.: at this step, the sample is stable 7 days at 2-8 C INJECTION : Inject 100 µl of the eluate into the chromatographic system. 5 25-OH VITAMIN D 3 FAST Warnings REAGENT F : TEST SOLUTION 25-OH VITAMIN D ng/ml REAGENT G : SERUM CALIBRATOR LYOPHIL. 25-OH VITAMIN D 3 Use and Reconstitution: Calibrators are used for calibration of the HPLC system. This lyophilised calibrator has to be prepared like a patient sample. Add exactly 5 ml HPLC water to the vial and mix for 15 min. When all material is dissolved, the solution is ready to use. Stability: 36 months if stored at - 20 C. After reconstitution the stability of the analytes is at least 7 days at 2-8 C and at least 1 month at -20 C. Store in a dark place. Don t use after expiry date. Packaging : 2 x 5 ml Warning: The calibrator derives from human serum, so it could be potentially infected. It must be handled with care. 200 ng/ml SPECTROPHOTOMETRIC DETECTOR PARAMETERS λ GAIN INTEGRATION TIME 265 nm 0,001 AUFS 10 sec. HPLC COLUMN PROTECTION To save the analytical column Reverse Phase PURSUIT XRs C18 50 x 4,6 mm, 3 µ, the use of Metasaver Precolumn Filter 0.5 um (1 x 10 pcs), cod. ZA6005 is obligatory. HPLC COLUMN CONDITIONING Install a new analytical C 18 Reversed-Phase column (PURSUIT XRs C18 50 x 4,6 mm, 3 µ). Disconnect the detector and filter 30 ml of H 2 O: Methanol ( 20 : 80 v/v ) solution and subsequently 30 ml of H 2 O per HPLC, set flow at 2 ml / min. Don t recycle the washing solutions. Filter the mobile phase with a vacuum system and a suitable filter of 0,22 µ. Condition the column with the mobile phase at a flow of 2 ml / min. and discharge the first 30 ml. Condition further on the column for 30 min. also at recycling phase. Finally inject the Chemical Standard and verify the quality of the HPLC run. It is possible to make analysis at recycling phase, providing that you filter the same mobile phase with a filter of 0,22 µ before any analytical run. If room temperature is > 20 C store the Mobile Phase at 2-8 C between an analytical session and another. COLUMN CLEANING Not necessary HPLC PARAMETERS LOOP 100 µl FLOW 2 ml/min.
