2 Features BGB Flash Cartridges are ultra-pure silica-based flash cartridges with tight particle size distribution and high surface area. Available in 50 µm and 30 µm particle size for difficult separation. Pressure-rated up to 27 bar (400 psi) for high flow rate and fast purification. Reduction of purification time by %. More concentrated fractions for easy evaporation and less solvent consumption. Features include 100 % guaranteed leak-free. Body in pharmaceutical-grade polypropylene. Closed-top spin-welded technology for stan dard hardware sizes. Pressure up to 27 bar (400 psi). Possibility of working at high flow rates for fast purifications. Available in 800 g, 1.6 kg and 3 kg formats. Luer and/or luer-lock connection for standard hardware types. Packed with ultra-pure silica-based material. Compatibility BGB Flash Cartridges are compatible with all instruments on the market: Agela, Analogix, Armen Instrument, Biotage, Büchi, ECOM, GE ÄKTA, Gilson, Grace, Interchim Puriflash, Teledyne Isco, Yamazen, etc. Different adapters are available. Please see on pages 10 and 11 or refer to Pressure limit BGB Flash Cartridges use a spin-welded technology that makes it possible to use the column up to 27 bar (400 psi) depending on column size. Please refer to the maximum pressure specified on each column and never exceed this value. Easy-Multi-Luer Outlet BGB Flash Cartridges in hardware W004 to W330 have a female luer-lock inlet and an Easy-Multi-Luer outlet (figure 2). To get a male luer tip, just remove the orange plug by pulling it down (figure 1). To have a luer-lock connection, simply unscrew the luer fitting by using the orange plug as a tool (figure 3). 3 Basic Information Report Sheet All boxes of BGB Flash Cartridges contain a report sheet. Every lot of filling material is tested to ensure good lot-to-lot reproducibility and the highest quality standard. Label Each BGB Flash Cartridge comes with a label including a full description of the phase with lot number and indicating the best parameters for column use as flow rate, maximum pressure and sample loading (figure 4). Storage before use Before use, store the cartridges in a dry place, away from light and dust, and at room temperature. The orange plugs must not be removed before the first use. Conditioning This stage is crucial to achieve the maximum performance of your column. It activates the silica and removes the air from the cartridge. For optimum purification, this step must be performed prior to the sample load. Rinse the column with 3 to 5 column volumes depending on the phase and the column size. Generally, the solvent (or mixture of solvents) for this step is the one used at the beginning of the purification. BE CAREFUL, and note that the use of a strong eluent for the conditioning will result in poor separation because of the high affinity of the solvent for the compounds to be purified. Depending of the solvent (or mixture of solvents) used, a normal heating phenomena can appear during this stage. To minimize this effect, and avoid a too much temperature increase, we strongly recommend to be at the lowest part of the flow rate range mentioned on the column. Drying the cartridge after the conditioning step will also result in a loss of column performance. Storage after use Figure 4 Normal-phase silica cartridges are single use columns and should not be stored or re-used. For the bonded phases C18, Cyano (CN), Diol (OH), Amino (NH 2 ) and HILIC please refer to the table below. Rinse 3 to 4 column volumes with the strongest solvent (100 %) to remove impurities. Rinse 6 to 8 column volumes with miscible solvents to reach at least 70 % ACN. Flush with 6 to 8 column volumes with miscible solvents to reach 100 % ACN. Please note that the orange plugs at the inlet and at the outlet of the cartridge must always be in place to avoid solvent evaporation. 3

10 Adapters for Large-Scale Cartridges Adapters for S0800, S1600 and S3000 Flash Column Hardware These adapters (stainless-steel luer-lock inlet and PEEK luer outlet) have to be used with BGB column hardware S800, S1600 and S3000 (please see page 5). They can be connected to any kind of luer connection. Luer-lock inlet: The stainless-steel inlet adapter is designed with a thread on half of the length and a smooth part on the other half. The thread is supposed to fit into the plastic when you screw in (figure 6). Stainless-Steel adapter for inlet port PEEK adapter for outlet port You have to screw it until the smooth part is engaged a few millimeters deep to insure a truly leak-free connection (figure 7). This can be installed by hand and needs to be screwed in manually. Luer outlet: The PEEK outlet adapter has to be firmly inserted into the column outlet to insure a truly leak-free connection (figure 9).
