4 Finnigan TSQ Quantum Discovery MAX H-SRM High Transmission at High Resolution Selective Reaction Monitoring (SRM) is a widely used technique for performing quantitative assays using a triple quadrupole mass spectrometer. Highly-Selective Reaction Monitoring (H-SRM) allows the user to have more stringent tolerance for precursor ion selection, which leads to higher analyte selectivity. This can lead to lower LOQs, better precision and accuracy at the limits of assay detection. Using H-SRM, the precursor ion is selected with a mass tolerance of ±.1 m/z or less, which dramatically increases analyte selectivity and reduces method development time when chemical noise is an issue. Consequently, this reduces the likelihood of false positives. Thermo s hyperbolic triple quadrupoles are the only triple quadrupoles currently on the market capable of such mass selectivity without a significant loss of sensitivity. H-SRM is a standard feature on all Finnigan TSQ Quantum instruments. Collision Cell The unique 9 collision cell design further reduces chemical noise by removing line-of-sight from the ion source to the detector. The 9 collision cell design also serves to reduce instrument foot-print, creating a conveniently sized instrument. reliability Elimination of Cross-Talk When performing LC-MS/MS analysis of multi-component mixtures by SRM, it is often necessary to use very short dwell times of 1 2 millisecond. In certain circumstances, for example in the environmental monitoring of pesticides, it is possible to get a large number of closely eluting compounds of the same chemical class, giving rise to the same product ions (see Table 1). In such assays it is possible that storage of the product ions can take place in the collision cell. In other words, fragment ions from one transition may still reside in the collision cell when the next SRM transition is monitored, resulting in cross-talk, and therefore incorrect quantification. The advanced design of the Finnigan TSQ Quantum collision cell results in the total absence of any cross-talk problems, as exemplified by the three closely eluting pesticides: triasulfuron, metasulfuron-methyl and chlorosulfuron (see Figure 1). 9 Application-Specific Software Xcalibur Data System Xcalibur is the most powerful and flexible MS data handling system available for Microsoft Windows. The software provides fully-automated control of the Finnigan TSQ Quantum products and Finnigan Surveyor LC, accelerating method development with intelligent optimization tools. The functional workspace-based instrument control view creates a simplified tune, calibrate, and compound optimization environment for analytical method development. Xcalibur controls LC gradient conditions, autosampler parameters, and PDA detection data as well as the Finnigan TSQ Quantum products. All complementary and confirmatory data are acquired and displayed in a single file. Automated Data Dependent experiments are routinely performed in which the instrument repetitively records MS spectra in a chromatographic run and switches to MS/MS mode only when a component actually elutes into the mass spectrometer. Thus, the maximum amount of information is generated in a single experiment. Xcalibur s homepage start-up screen offers easy navigation through the process of Instrument Setup, Sequence Setup, and Data Acquisition. Review of results is simple with three interactive, applicationspecific browsers: Qual, Quan, and Library Browsers. LCQUAN Software Quantitation Software for Regulated Laboratories LCQUAN has been expanded into a secure data system for quantitative analysis.
