Hardware Designed for Optimum Performance and Service Life High efficiencies and negligible extra-column effects are a certainty because Higgins Analytical columns use high quality PEEK and titanium frits that are only 0.75mm thick. Frit porosity also plays an important role in HPLC column design. Many manufacturers use 2µm porosity frits with their 5µm sorbents making high sensitivity work problematic since even the best particlesized 5µm materials can have 1% or more of sub 2µm material. These minute particles can pass through the frit and into the detector causing spiking and baseline noise. 7 CLIPEUS CLIPEUS Offers a Wide Range of Hydrophobicity The highly pure silica backbone and the thorough end-capping process that are characteristics of CLIPEUS HPLC columns enable a wide choice of column selectivity without compromising chromatographic performance. Not only do the columns with relatively small cyano and phenyl functional groups perform as efficiently as do the longer chain C8 and C18 versions of CLIPEUS, they also produce symmetrical peaks for acidic and basic analytes. This symmetrical version (P/N HK-GUARD-FF) is called for when Heat Transfer Characteristics The thermal conductivity of the plastic materials in Higgins Analytical s cartridge holders (~0.15 BTU/(hr)(ft 2 )(ºF/ft) is one to two orders of magnitude smaller than aluminum (113) and stainless steel (~1,000) commonly used in other manufacturer s cartridge holders. Thermal equilibration rate measurements were made to determine how these differences in thermal conductivity would effect re-equilibration time after a temperature change. Experimental Results Relative temperature equilibration times for a Brownlee MPLC Holder and a Higgins HAIPEEK holder were determined chromatographically. A 40x3.2mm HAISIL 100 C18 5mm cartridge (P/N HM185- HF0432) was used for the analysis of fluorene in a 70% acetonitrile/water mobile phase at 420mL/min in both holder types. After determining the analyte s retention time at room temperature (21.7ºC), the assembly was placed in a forced air oven set to 35ºC. Retention time measurements were made at approximately two minute intervals until a steady state was reached. The figure to the right shows that rates for both holders were very similar, with equilibration being reached in a little less than 10 minutes. Notes Retention time precision was approximately minutes for these data. Oven temperature was measured to 0.1ºC. Oven temperature was 35.9ºC during the Higgins holder experiment. Temperature was 35.0ºC during the Brownlee holder experiment. Visual Contamination Checks One of the biggest issues with guard column use is the uncertainty about when it should be changed. A recently published survey indicated that the typical user changes a guard column very few times throughout the analytical column s lifetime. One reason for this poor practice is that, except for back pressure build up, most indications of guard column failure come long after the analytical column has been damaged. With a Guard-Disc, a more rational approach can be taken. The Guard-Disc s unique design makes it the only HPLC column guard system that is capable of being visually inspected on a periodic basis. The Holder can be quickly opened to inspect the top surface of the disc for contamination, particles, or discoloration. Symmetrical Version for strength and flexibility. DMSO, Me 2 Cl 2, and THF can cause PEEK to swell over a period of time, so they are not recommended for routine use. The Disc s characteristics are outlined in the table below. The C18B discs exhibit slightly chromatographic performance than the C18A type, but they should not be used with chlorinated solvents. Volume Variance: The Holder and Disc s contribution to an HPLC system s total system volume variance is about 20mL 2 at 200mL/min, so under isocratic conditions, some performance loss can be experienced with columns with I.D. s smaller than 4.6mm.
