1 ACETONE IN URINE BY GC/MS Headspace Code GC10110 BIOCHEMISTRY The acetone or dimethyl ketone is a product which falls into the biochemical physiological small amounts are formed as a product of the metabolism of fats and carbohydrates. Acetone is used in the laboratory as organic solvent and drying of glassware as an adjuvant laboratory and is used in cosmetics to remove the glaze. In the metalworking industry as acetone is used as a solvent in paints, glues, dyes, etc. The absorption of acetone takes place mainly through the respiratory system and to a lesser extent, through the skin so it can be irritating to skin and mucosa. Its power is very limited systemic toxicity. Exposure to high concentrations may cause dizziness, nausea, vomiting under rapidly reversible after exposure ceased. The proposed method determines the acetone injected into GC-MS / headspace of a rate of urine sample earlier in appropriate solvents. The levels of acetone are determined on a urine sample at the beginning and end of shift. 2 TECHNICAL FEATURES Principle of the Method : This method allows to determine the urinary acetone by GC-MS / headspace.the acetone is first diluted and, after the addition of a stabilizer, is directly injected into GC. Recovery : 99% Sensitivity : about 0,01 mg/l by MS Dynamic Range: 0,01-10,0 mg/l by MS Normal Range: 0,10 2,0 mg/l Components of the kit : All reagents are ready to use and stable 3 years at 2 8 C. Reagent A Diluting Solution, 1 x 200 ml Reagent B Test Solution 1,6 mg/l, 1 x 20 ml Minimum Instrumental equipment required: GC/MS Operational Computer Optional Equipment: Autosampler Urine Collection Procedure: After work shift, collect 10 ml of urine in a suitable test tube. Stable about 60 days at 20 C. 2
3 PREANALYTICAL PROCEDURE Preparation of Test Solution STEP 1 : Dispense in a tube: 1800 µl of Reagent A Diluting Solution 200 µl of Reagent B Test Solution Inject 1000 µl of solution into gas chromatograph GC. Verify that the Test solution has retention time similar to fig. 1. If the Test is all right you can start with the analytical procedure; if not, check the functionality of the analytical system. ANALYTICAL PROCEDURE STEP 1 : Preparation of Calibration Standard Solution at 0,16 mg/l Dispense in a tube: 1800 µl of Urine free from Acetone 200 µl of Reagent B Chemical Standard IMPORTANT: This Calibration Standard Solution is prepared from time to time To verify that the urine is effectively free from Acetone, to prepare a Blank: 1800 µl of Urine 200 µl of H 2 O HPLC grade If the Blank urine presented the peak of Acetone, proceed to identify the quantitative difference between the areas / heights of the peaks of Acetone in urine enriched and blank. The samples are directly injected transfer 2 ml in GC vials. Close vials hermetically Mix at Vortex Place them in the Sampler Prepare work list and start the serie according to the system s specifications Release N 002 Acetone in urine by GC/MS-headspace February

Do not condition column/s if connected to the Massa Detector. CLEANING OF THE COLUMN Disconnect the detector. Keep the column at the highest temperature for the recommended time.
