13 Entrance test Before student can think of own method he/she has to have knowledge gained at previous levels of studies. To chek, repeat and prepare oneself he/she will do: Quiz with answers and direction to literature references To check understanding, application, analysis of the process in instrument This has to be done before start of the method development plan Will be on Moodle platform
14 Question data base has to be made for topics, appropriate for the highest level of education: 1. How GC works Parts Principle Retention times Columns Injectors (liners, septa, peak focusing) Detectors 2. How MS works EI, CI Data aquisition SIM, SCAN Data analysis- qualitative and quantitative analysis (IS, external standars, standard addition) Check is it working in a proper way 3. Sample prep Extraction Clean up
15 Examples of questions Instead of the question: What is the retention time? they will be asked to read and compare RT from real chromatograms. How is RT influenced by the temperature and or gas flow? they will be asked to explain why some changes in real chromatograms happen due to temperature and flow changes. To explain CI and EI they will be asked to gues which one type of ionisation is used to get certain spectra and how they can increase sensitivity of their instrument.... Thus we will encourage both analytical and synthetical way of thinking as well as evaluation before they get main task.

21 Real, problem based example-test on their own substance Evaluation of the developed method for benzophenone analysis in water Internal standard method, benzophenone d-5 Extraction by SPE procedure, OASIS HLB 60mg Evaporation to dryness and derivatisation with PFBHA The results of the testing method for benzophenone analysis are given in Test excell file with four sheets: Calibration, Blank LOD LOQ, Precision, Recovery.
22 Calibration Worksheet Calibration contains the surfaces of target ions peaks for benzophenone (BP), m/z 377 and IS, (BP-d 5 ), m/z 382. Calibration has been done 3 times for concentration levels of 20, 100, 200, 400, 750 and 1100 ng/l. Concentration of IS was 500 ng/l. Task: present your plan of making solutions if you want to do SPE with 200 ml of water. Task: Calculate average linear equation for calibration curve and coresponding coefficient of correlation.

24 LOD and LOQ In worksheet named Blank LODLOQ data for 7 measurements of blank samples (water matrix spiked with internal standard in same concentration as calibration standards) are given. Task: Calculate LOD and LOQ
25 Precision Instrument precision was checked by injecting calibration standard at 6th concentration level (1100 ng/l) three times from the same vial. Task: report on precision of those injections by analysing responses of the detector for target ion of BP Method precision was checked by analysing injections in triplicate for two different concentrations within calibration curve (e.g. At 200 and 750 ng/l level). Task: report on precision of those injections by analysing ratio of responses of the detector for target ion if BP and target ion of IS for triplicate measruerments at each concentration level
