5 Introduction to Research Projects Chemistry, as a blooming and enabling science, is always at the focal point of every important modern societal challenge. Cutting-edge, multidisciplinary research issues are being explored along with traditional disciplinary research topics. My research experience embraces analytical chemistry, biosensors, physical chemistry, and exposure to biochemistry and material chemistry. The following describes some of my research projects. SIEP-based Electrochemical Approach for mirna detection. MiR-27a, a key biomarker of breast cancer, was taken as a target model to access the detection ability of the method. A hairpin probe (HP) hybrids specifically with mir-27a was designed and modified on MB (HP-MB), which was acted as an isolation vehicle. SIEP amplifications were performed at the ends of oligonucleotide primers, which were loaded on PS. In this project, I investigated and proposed the possible action and interaction of mir-27a with hairpin probe according to its properties. Different experimental procedures, such as electrophoresis, were indentified and conducted to monitor the incubation condition. The possible mechanism for the amplification of the signals was finally concluded. This research project is currently at the stage of conducting animal tests. After passing the animal test, the thesis will be submitted to Analytical Chemistry (journal). Modified electrode surface prepared by electro-grafting to immobilize enzymes and its applications in drug screening on the platform of microchips. In this research, different methods were investigated to enhance the detection sensitivity and selectivity of electrode by electro-grafting an organic layer such as those with amine, carboxyl and alcohol group. One discovery is that when grafting a layer of polyoxyethylene on glassy carbon electrode, a layer of high water affinity can be formed. The structure of polyoxyethylene can prevent the adsorption of protein on the electrode and maintain its sensitivity. By controlling the electric potential, the modified and de-modified of electrode can be controlled in practice. This technique is of great potential usage in pharmacological area. Working with principal investigators, I proposed different compound for electrode grafting to modify the electrode so as to fit its application, which was recognized as one of the milestones of this project. In proposing the possible method, several factors should be considered, including its structure and possible interaction with the electrode, different drugs, and enzymes. Catalytic conversion of C2-4 molecules to polycyclic aromatic hydrocarbons over Silicon Carbide(SiC) a proposed way of formation of PAH in the universe. In this research, possible catalyst SiC present in the universe in the action of converting C2-4 molecules into PAH of different sizes and the mechanism in the process were investigated. The first step is the confirmation of SiC as a possible catalyst in the conversion of C2-4 hydrocarbon into PAH. The effort is then to figure out the mechanism behind. Gas phase hydrocarbon with different sizes and saturation is experimentally allowed to pass over SiC in order to investigate the proposed pathway. Experiments on propane, propylene, butylene, butane, propyne and CO are carried out afterwards. In this project, I proposed the possible mechanism and the way of confirmation. After that, I worked with the research team together in setting up the reaction chamber and monitoring the condition. The experiment and the analysis are conveyed that a crucial practical aspect would be the alignment in different parts of the set up; if the alignment is not in good condition, a great deviation of result would occur lowering the consistency, in which adjusting the alignment needs to be highly corporate and meticulous.
