1 HPLC Column Material - Bulk Ware Constantly growing demands on separation efficiency and availability of solid phase material for packing HPLC columns and column packing stations have motivated us to provide a wide range of separation materials. The availability of a many different void sizes allows various applications with optimized selectivity and capacity. The uniform distribution of particle sizes crafts the best compromise between performance, operating pressure and processing time, thus creating a good price-performance ratio. All spherical silica gels are mechanically very robust. Therefore, they can be packed several times into analytical or preparative HPLC columns. Additionally, each production group passes through an extensive quality control and testing process to ensure consistently good quality for each production batch. Spherical silica GEL column material is manufactured at very high chemical purity standards. Organic and trace metal content are controlled within very strict limits. Reproducible separation results, direct scale-up without time-consuming method redevelopment, are direct results of tight manufacturing parameters such as chemical purity, surface area, void volume and void size. 2 Spherical Silica GEL SMB (Super Micro Bead) The "Super Micro" spherical silica gels are ideally suited for normal phase chromatography, for both analytical and preparative HPLC. For analytical HPLC, we offer a selection of 3 to 5 µm. The 5 µm particles are the recommended particle size for method development and also often help optimizing the method. For preparative HPLC, in pilot plant or for production processes, the 10 µm column materials are preferably used. The 15 µm versions offer a good compromise between performance and costs. The particle sizes of 20 and 20/45 µm are well suited for medium HPLC columns. Column material with void sizes of 7, 10 and 20 nm are recommended for cleaning small, natural and synthetic molecules, lipid resolvable organic matter and small organic chemical compositions. Larger particles of 30, 50, 80, or 100 nm are preferably used for optimizing the capacity and the yield of the purification of a medium with large peptides and proteins. 3 Spherical Silica GEL MB (Micro Bead)) Micro-spherical silica gels are ideally suited for the normal phase chromatography, flash chromatography and for the adsorption chromatography. Especially in flash chromatography a good quality of the silica gel is critical in order to achieve optimum performance. The distribution of the particle sizes and the particle shape determine the purification and the speed. For efficient mass transfer spherical silica gels have a tight control of particle size distribution, which allows less than +/- 10% for the defined distribution. Two standard distributions of particle sizes are available Surface, water content and surface ph value determines the capacity of the silica gel. This column material is totally porous silica gel, and the strict control of void volume and void size distribution ensures the accessibility for the target molecule. Silica gels with void sizes of 7 and 10 nm are recommended for cleaning small, natural and synthetic molecules, lipid resolvable organic matter and small organic chemical compositions. 4 Granular Silica GEL The granular silica gels are the most popular area for normal adsorption chromatography. The production of many 1,000 tons of basic material each year for the subsequent production of silica gels ensures a high level of consistency and reproducibility. In addition, production in such a large scale allows optimum process control for excellent quality and a very good price-performance ratio. The granular silica gels are manufactured to match very high chemical purity standards. Organic and trace metal content are controlled within strict limits. Reproducible separation results, direct scale-up without timeconsuming method redevelopment, are the direct result of tight manufacturing parameters such as chemical purity, surface area, void volume and void size. Granular silica gels are particularly well adapted to applications that do not require a high column bed plate number, e. g. open columns, or LC columns and even for a group process. 5 Bonded Silica GEL All of these silica gels can be derivatized to form bonded silica gel for the "Reverse Phase Chromatography" or for some special chromatography methods. Derivatization The growing interest in complicated synthetic or recombinant peptides has expanded and the demand for high-performance bonded silica gel has vastly grown. The usage of bonded phases for injectable biological products has also created new demands. Optimized Quality The fact that the starting gels are produced in very large quantities makes an important contribution to the overall reproducibility of these reverse phase silica gels. Each production lot is subjected to extensive testing on carbon loading, chromatography retention time, efficiency and peak symmetry. The quality of the surface finish is also checked. Optimized Chemistry The silica gels are made with mono-functional organosilanes and undergo end capping surface treatment. TRI-functional with or without end capping can be produced on request. The quality of the surface treatment is assessed by the test of the phenol/pyridine to detect any residual free acid OH group on the silica surface. Overview of column material versions for packing chromatography columns. C18 Silica Gel This is the most versatile and popular reverse phase. The void size must be chosen carefully and arises from the molecular weight of the molecule. C8 Silica Gel Universal yet less powerful than the C18, it is the primary choice for investigations. Phenyl Specific reverse phase for the medium / polar molecules, shows high selectivity for acceptor/donor compositions. C4 Silica Gel Extremely powerful when bonded, in large void size silica gel for bio-chromatography and polar compositions. NH2 The amino-silica gels separate a variety of sugars with mobile phase from acetonitrile/water. Alternatively, it functions with hexane / ethyl acetate as a 'less active' mobile phase. In addition, since the surface contains primary amino groups, NH2 can be used as a base for the coupling of chiral ligands. CN Can be used both in normal phase and in reverse phase chromatography, for steroids and catechols. Diol Especially for aqueous GPC applications diol can be used for normal phase chromatography, for example for the separation of phenol. 5/5
