2 3M Food Safety
3M Petrifilm Salmonella Express System 3M Petrifilm Salmonella Express System is a qualitative test used for the rapid detection and biochemical confirmation of Salmonella in enriched foods and food process environmental samples, replacing traditional agar methods. o Sample ready media: 3M Petrifilm technology o Easy to use o No special equipment required o Allows for greater uniformity of results vs. dehydrated agar o Longer shelf life compared to agar plates o Compact/stackable plate & disk reduce waste 3 o All-in-one test method: provides presumptive and biochemical confirmed results o Quicker time to biochemical confirmed result (44-52 hours) o Confirms all circled presumptive positive colonies on plate all at the same time
3M Petrifilm Salmonella Express System Method Overview Two Methods: High microbial background vs. Low microbial background o Both methods use 3M Salmonella Enrichment Base + 3M Salmonella Enrichment Supplement o Only high microbial background method uses R-V R10 as secondary selective enrichment o Single streak method for plating o Single incubation temperature of 41.5 ± 1 C throughout method o Flexible range of incubation time to support laboratory flexibility 4
3M Petrifilm Salmonella Express System Matrices Evaluated in Method Validation High Microbial Load >10 4 CFUs Raw Meats Ground chicken Ground beef Ground pork Seafood Frozen, uncooked shrimp Fresh Produce Spinach Low Microbial Load <10 4 CFUs Processed Meats Cooked chicken nuggets Egg Products Pasteurized, whole liquid eggs Pet Food Dry dog food Environmental Stainless Steel 5
6 3M Food Safety 3M Petrifilm Salmonella Express System Procedure Media Preparation & Enrichment Procedure
3M Petrifilm Salmonella Express Plate Hydration Procedure o Hydration Diluent = reverse osmosis water, distilled water or Butterfield s Phosphate Diluent 7
3M Petrifilm Salmonella Express Plate Qualifying Plate: Hydrated Plate Color Card o o o A Hydrated Plate Color Card will be placed in each Box and Case of the 3M Petrifilm Salmonella Express Plates. This is a great tool for incoming inspection to quickly check if the plate is acceptable for use. If Hydrated Gel is Red or Bright Pink, plates are acceptable for use. If the Hydrated Gel is orange or brown, plates should NOT be used for testing due to improper storage or transit (extended exposure to high temperatures). Hydrated Gel Area 8

Identifying Salmonella Species The following are presumptive positive Salmonella species: The following are Non- Salmonella species Blue colonies, green colonies, blue to green colonies, and/or black colonies with or with out a yellow zone and/or associated gas bubble Red, dark red and brown colonies with out a yellow zone and/or associated gas bubble Red, dark red and brown colonies with a magenta zone 10 Must circle presumptive positive Salmonella colonies prior to disking

12 3M Food Safety Plate Sample Presumptive Positive & Biochemical Confirmation
3M Petrifilm Salmonella Express System Procedure Biochemical Confirmation The biochemical confirmation result of the 3M Petrifilm Salmonella Express System is statistically equivalent to the biochemical results of the reference methods. What does it compare to? Standard method testing through biochemical identification (Vitek or API). Not equivalent to O&H serological results. 13
3M Petrifilm Salmonella Express System Procedure A Comparative Evaluation of the 3M Petrifilm Salmonella Express System for the Detection of Salmonella species in Food and Environmental Surfaces AOAC Method Validation Testing conducted by third party reference lab Comparison testing to reference methods: USDA/FSIS-MLG 4.05 (2011) Isolation and Identification of Salmonella from Meat, Poultry, Pasteurized Egg and Catfish Products FDA/BAM Chapter 5 Salmonella method ISO 6579: Microbiology of food and animal feeding stuffs-horizontal method for the detection of Salmonella spp. 14

3M Petrifilm Salmonella Express System Procedure Summary of Results - 3M Petrifilm Salmonella Express System Performance No statistically significant difference in the number of positive samples detected by the 3M Petrifilm Salmonella Express System and the reference methods for all matrices analyzed. 17 Summary from 3 rd Party Reference Lab The 3M Petrifilm Salmonella Express System demonstrated reliability as an alternative qualitative identification method for the detection and biochemical confirmation of Salmonella in select foods and environmental surface.
3M Petrifilm Salmonella Express System Validation: 3 rd Party Reference Lab Summary o o o o o For all matrices tested, the 3M Petrifilm Salmonella Express method was equivalent to the reference methods There were no false positive results Easy to Streak Like Single Streak Method Easy to interpret High background foods were easier to interpret on the 3M Petrifilm Salmonella Plate compared to the Reference Method Faster time to result Like the [3M Petrifilm Salmonella Express] Confirmation Disk compared to the Reference method This is a great product and the 3M team should be very proud --Pat Bird, Study Coordinator, Q lab, USA 18
